GenoMatrix完成对马属生育力候选基因SPATA1的全外显子变异独立验证报告。该研究对SPATA1基因的63个变异位点进行了系统性审查,发现并修正了32%的注释错误。
其中最关键的发现是chr5:76,147,665 G>A位点的注释纠正。该变异原被标注为stop_gained(功能丧失),但经独立验证后确认为missense突变(S206L)——即氨基酸替换而非蛋白质截断。这一差异对临床解读具有重大影响。
需要指出的是,32%的错误率仅限于SPATA1基因的本次验证范围,不代表全局注释错误率。
验证过程中还发现了VEP(Variant Effect Predictor)在负链基因注释中的系统性偏差。GenoMatrix已建立负链基因变异的交叉验证流程,以减少此类错误。
GenoMatrix has completed an independent verification report on the fertility candidate gene SPATA1 in equids. The study systematically reviewed 63 variant sites in the SPATA1 gene and identified/corrected 32% annotation errors.
The most critical finding was the annotation correction at chr5:76,147,665 G>A. This variant was originally labeled as stop_gained (loss of function) but was independently verified as a missense mutation (S206L) — an amino acid substitution rather than a protein truncation. This difference has significant implications for clinical interpretation.
It should be noted that the 32% error rate is limited to the SPATA1 gene verification scope and does not represent the global annotation error rate.
The verification process also revealed systematic biases in VEP (Variant Effect Predictor) annotations for negative-strand genes. GenoMatrix has established a cross-validation workflow for negative-strand gene variants.
